Description 

Bremelanotide (PT–141) is a synthetic cyclic heptapeptide analog of a–melanocyte-stimulating hormone (a-MSH). It is a potent agonist of melanocortin receptors, particularly MC3R and MC4R. 

Mechanism of Action 

PT-141 selectively activates melanocortin receptors in the central nervous system (hypothalamus and limbic system). This triggers: 

• Increased sexual arousal and desire via neural pathways independent of vascular mechanisms 
• Modulation of dopamine and other neurotransmitters involved in sexual motivation

Indications & Usage

Contraindications & Warnings 

Do not use if you have: 

• Known hypersensitivity to Bremelanotide or any excipient

• History of melanoma or skin cancer 
• Pregnancy or breastfeeding 
• Age under 18 years 

Warnings 

• Use only under medical supervision. 
• Not a substitute for addressing underlying relationship, psychological, or medical causes of sexual dysfunction. 
• May cause temporary increase in skin pigmentation or darkening of moles. 
• Discontinue immediately and seek medical attention if severe symptoms occur (e.g., severe nausea, high blood pressure, or allergic reaction). 

Dosage & Administration 

• Subcutaneous injection only. 
• Standard / Effective Dose: 1.0-1.75 mg (24-42 clicks) 
• Starting / Low Dose: 0.5-1.0 mg (12-24 clicks) – recommended for first-time use 
• Frequency: As-needed, at least 45 minutes (ideally 45-60 minutes) before activity 
• Maximum: Do not exceed 1 dose per 24 hours or 8 doses per month 
• Timing: Inject 45-60 minutes prior to anticipated activity 

Administration Instructions 

1. Attach a sterile needle. 
2. Dial your dose (e.g., 24 clicks = 1.0 mg). 
3. Clean injection site (abdomen or thigh) with alcohol swab. 
4. Insert needle at 45-90° angle and inject slowly. 
5. Withdraw needle, dispose safely, and cap the pen. 
6. Rotate injection sites. 
7. Always inspect solution: Do not inject if cloudy, discoloured, or containing particles. 

Side Effects 

Common or potential reactions include: 

• Mild to moderate nausea (most common, usually temporary) 
• Flushing, headache, or light-headedness 
• Injection-site irritation or redness 
• Temporary increase in skin pigmentation or darkening of freckles/moles 
• Rare: Increased blood pressure, vomiting, or fatigue 
• If severe nausea, vomiting, or cardiovascular symptoms occur, discontinue use and seek medical attention. 

Storage & Handling 

• Store either in refrigerator (fridge); never freeze or in a cool dry place (≤25-30°C / 77-86°F). Keep pen capped and protected from light and heat. 
• After first use: Same storage conditions. 
• Keep out of reach of children. 
• Discard if expired or solution appears abnormal.
